Induced hyperglycaemia (oral challenge)

Purpose

Investigation of pancreatic islets of Langerhans beta-cells. Inhibition of growth hormone secretion (GH).

Subject preparation

Fasting overnight.

Product

Glucose.

Dose
  • Adults: 75 g dissolved in 250 ml water, – oral administration.
  • Children: 1.75 g glucose/kg body weight (do not exceed 75 g).
Protocol
  • Sample at T0 (1 fluoride or iodo-acetate tube, 2 dry tubes).
  • Oral administration of the correct dose of glucose in 250 ml water.
  • Samples (1 fluoride or iodo-acetate tube and 2 dry tubes) T+30, T+60, T+90, T+120, T+150 and T+180 min after ingestion of the glucose.
Samples

Immediately cold-centrifuge the blood samples taken at each time point. Freeze the serum (for the assay of GH, insulin and/or peptide C) and store the plasma (for the glycaemia) at +4°C.
