Clinical significance
The C1 component of complement is a macromolecular complex of 2 C1q sub-units and a tetramer comprising 2 C1r and 2 C1s sub-units. C1q is recognized by classic pathway activators. Components C1, C4 and C2 interact to form the C3 convertase (C4b,2a) of the classic complement activation pathway. C1 activation is regulated by an inhibitor (C1 inhibitor).Activation of the classic pathway is associated with decreased responses in the CH50 assay and reductions in the levels of components C3, C4, C1 and C2; this can be associated with glomerular disease, endocarditis and systemic lupus erythematosus.
