Clinical significance

Synthesized by the reticuloendothelial system, especially in the liver where its production is not vitamin K-dependent, and by platelets (20% of the circulating pool). Acquired Factor V deficiency can occur in severe liver disease such as cirrhosis or chronic hepatitis, fibrinolysis, diffuse intravascular coagulation and rarely neoplasia associated with the production of anti-Factor V antibodies. It is a mutation in the gene encoding Factor V (the Leiden mutation) which underlies the phenomenon of activated Protein C resistance.
