Clinical significance
HE4 is a protein that is over-expressed in patients with ovarian cancer, even in initial stages (I and II) of the cancer, mainly in serous cancers. Its expression is independent of that of CA125 and occurs in 50% of cancers that do not express CA125. The combination of these two serum markers improves the sensitivity and specificity of the detection of ovarian cancers at early stages and their relapse. The ROMA algorithm evaluates a risk of malignancy by combining serum concentration measurements of HE4, CA125 and menopausal status. It allows the classification of patients according to level of risk of malignancy, low or high, by taking into account their menopausal status.
Further information
For the ROMA algorithm, HE4 and CA125 are assayed with the same technology, which does not authorise integration of results sent for CA125.
