Prekallikrein is a zymogen which, when acted upon by Factor XII or XIIa, forms the serine protease, plasma kallikrein which cleaves high molecular weight kininogen to form bradykinin. Prekallikrein is involved in the initial, "contact phase" of the intrinsic process of blood coagulation.This "contact phase" does not seem to be crucial in physiological terms because deficiencies in either Factor XII, high molecular weight kininogen or prekallikrein are not associated with any kind of bleeding disorder.
